Brief summary of research:
The central theme in the de Haan research group is the interaction between viruses and their hosts at the molecular/cell biological level as well as at the level of the organism. These virus-host interactions are studied for coronavirus, influenza A virus and respiratory syncytial virus, which are important infectious agents of animals and man.   

The de Haan group studies replication of viruses in cells as well as in the whole organism. On the one hand, his group developed pioneering techniques such as bioluminescence imaging of coronavirus infections to study the spread of infection in living mice, while on the other hand state-of-the-art cell biological techniques are being used, such as live cell imaging, for example to study the biogenesis and dynamics of coronavirus replication complexes in the cytoplasm of the host cell. 

His group is furthermore interested in virus-cell attachment and subsequent endocytic uptake of virus particles. For example, novel systems were developed for the expression of recombinant soluble influenza glycoproteins. The recombinant proteins are employed to study the interactions of influenza viruses derived from different host species with sialylated cellular receptors among others by using glycan arrays. Furthermore, the influenza A virus group focuses on the endocytic uptake of IAV particles. They were the first to show that influenza virus particles are efficiently taken up by cells via macropinocytosis.

In addition, the de Haan group is involved in the development of innovative vaccines against influenza A virus and respiratory syncytial virus based on our recombinant protein technology. The vaccine potential of recombinant soluble hemagglutinin and neuraminidase proteins of influenza virus was recently demonstrated using mice, chickens, ferrets and pigs. Similar approaches are being applied to develop a vaccine to protect against respiratory syncytial virus.

Awards: Xander de Haan was awarded with an NWO VIDI (2004) and with an Utrecht University High Potential grant (2006, together with dr. Fulvio Reggiori). In 2006 he received the KNAW Beijerinck Premium. PhD students working for Xander de Haan have been awarded with the H.G.K. Westenbrink price of the Stichting Stimulering Biochemie Nederland for best PhD-thesis in the area of biochemistry and molecular biology (2010; Matthijs Raaben) and with the Westerdijk Award of the Koninklijke Nederlandse Vereniging voor Microbiologie for the best PhD-thesis in the field of (medical) microbiology or infectious diseases (2012; Marne Hagemeijer).
