Profile

Willem van Eden (MD, PhD) is immunologist and medical microbiologist. He was a PhD student in the dept. of Immuno-hematology  (Jon J van Rood) at Leyden University and he received in 1983 a prestigious prize from the Royal Society of England (Bruno Mendel Award) which made it possible for him to stay for two years in the group of Irun Cohen at the Weizmann Institute of Science, Rehovot, Israel, where he worked on experimental autoimmunity.  In 1984 he received the Prix Zambon Benelux for his work in the area of infectious disease pathology.

Soon after his return from Israel van Eden attracted attention because of two simultaneous publications in the Lancet. This was a trigger for the daily newspaper Volkskrant to highlight the work of van Eden concerning the relationship between infectious diseases and autoimmune diseases.

In 1988 van Eden and colleagues discovered the role of stress proteins in self-tolerance. This serendipitous finding was published in Nature and was for two years in the top-100 of mostly cited paper in the biomedical domain. Resulting from this original finding are still current activities of van Eden and his group directed towards development of an immuno-modulatory vaccine against chronic inflammatory diseases on the basis of heat shock proteins. Van Eden is scientific director of Trajectum Pharma, which is a University of Utrecht derived spin-off company.

Van Eden has coordinated 4 EU consortia.

The activities of van Eden are in both the veterinary and the human domain. He is a member of the Dutch Health Council (National Vaccination Program) and the Council Group on Infection and Immunity.  He is a member of EVAG, the standing committee of European Vaccine Policies of the European Centre for Disease Prevention and Control (ECDC).

1) TOLERAGE

Normalisation of immune reactivity in old age – from basic mechanisms to clinical application

When restoration of the immune response in older age is the target of basic and clinical research, boosting innate and adaptive immunity, e.g. by more efficient vaccination strategies, is the general approach. However, TOLERAGE has a fundamentally different focus, i.e. restoring the age-dependent decline of auto-tolerance that underlies the development of the paradigmatic age-associated diseases atherosclerosis (AS) and rheumatoid arthritis (RA). These diseases represent the major medical and socioeconomic burden in aging societies. They start earlier in life but become clinically manifest with increasing age. To reach this aim a consortium of ten partner groups was formed that have an outstanding track record in areas of immunology pertinent for this task. The work of TOLERAGE will focus on three major issues, viz.
  1. Investigating basic principles of the induction of central and peripheral tolerance in mice against autoantigens, including heat shock protein 60 (HSP60) that has been shown to play a major role in the development of AS and RA.
  2. Determination of the mediators of central and peripheral tolerance that are responsible for the T effector/T regulatory cell dysbalance in older age in general and in AS and RA in particular.

Translation of the results obtained in (a) and (b) into practical application for the prevention and treatment of AS and RA by appropriate vaccines. For all partners, important technical platforms will be available, including unique transgenic and knockout mouse strains, sophisticated models for in vitro and in vivo transfection of dendritic cells (DCs) and a well established Functional Genomics Facility.

2) Advanced Immunization Technologies (ADITEC)

Aditec is a collaborative research programme that aims to accelerate the development of novel and powerful immunisation technologies for the next generation of human vaccines. Scientists from 13 countries and 42 research partners collaborate in the Aditec project.
Aditec comprises some of the most competitive European research groups from universities, research institutions and biotech companies together with top US groups on systems biology and adjuvants. Different aspects of vaccination, from basic research, new technologies to clinical trials and public health, are covered.
Aditec started in October 2011, is a 5-year project and is funded by the EU 7th Framework Programme.

 

3) BioVacSafe

Summary

Since their discovery, vaccines have protected millions of people worldwide from a broad range of infectious diseases, making them one of the most effective public health interventions out. New and better vaccines are still urgently needed, yet their introduction is hampered by lengthy and expensive vaccine safety testing procedures. The aim of the IMI-funded BIOVACSAFE project is to develop cutting edge tools to speed up and improve the testing and monitoring of vaccine safety, both before and after release to the market. By bringing together Europe’s top industrial and academic teams for the first time, the project will ultimately usher in a new generation of safer, more effective vaccines.

Therapeutic modalities based on macromolecules of biological origin such as proteins, peptides and oligonucleotides have a huge pharmacological potential due to their highly selective mode of action and their potential activity against targets that are considered ‘non-druggable’ by more traditional small organic molecules.

However, the widespread application of many potential macromolecular therapeutics has been very limited due to pharmacokinetic and drug disposition limitations at both the tissue and cellular level. Therefore, their success in developing future innovative medicines will heavily depend on improvements in both chemistries and delivery technologies that can address these limitations.

Therefore leading European experts from sixteen academic research institutions and SME as well as seven pharmaceutical industry research organizations in the area of delivery and uptake of biological macromolecules officially launched the COMPACT project on November 1st, 2011.

The close cooperation of academic teams, pharmaceutical companies and Small to Medium Enterprises (SMEs) in the COMPACT project will provide a unique platform of expertise of an EU-supported public-private partnership to persistently improve industrial competitiveness and Public Health in Europe.

The COMPACT proposal has been published in the fourth call for proposal of the Innovative Medicines Initiative (IMI), which is part of the EU Seventh Framework programme (FP7).
