Science. Throughout my scientific career I have worked mainly on structural characterization of membrane-associated and processes and those carried out by transient soluble assemblies. The work includes both development of computational methods (e.g., cryo subtomogram analysis, integrative modeling) and experimental studies of the eukaryotic protein translation, modification, and degradation machinery. A strong computational basis puts me in the unique position to address structural biology questions from unusual angles at the interface of molecular and cellular structural biology.

Most important achievements include (i) developing the first method for subtomogram averaging (Förster et al, PNAS 2005), which is indispensable for in situ structural biology using CET, (ii) achieving subnanometer resolution for the in situ structure of an asymmetrical complex (ER-associated ribosome) (Pfeffer et al, Nat. Comms. 2015), which structurally differs from detergent-solubilized structures highlighting the importance of studying membrane protein complexes in their native settings, and (iii) determining the first atomic structure of the complete 26S proteasome by integrative structure determination (Beck et al, PNAS 2012).

After successfully establishing an independent research group that transforms computational developments into unique structural and functional insights into protein translocation and degradation, I have been appointed as full professor at Utrecht University in 2016.

Complexes that are associated to lipid membranes and transient macromolecular complexes carry out many important cellular functions, but their structures are largely elusive to date because these types of macromolecules are extremely difficult to assess by established reductionist approaches. We study such complexes in situ using cryo-electron microscopy techniques, protein-protein interactions, and computational modeling. Using this approach, we elucidate the molecular machinery that is associated to the endoplasmic reticulum (ER) and mitochondrial membranes. Specifically, we are interested in the structural basis of ER- and mitochondria-associated protein biogenesis and degradation and in the development of the required methodology.
